SIP를 이용한 난배양성 미생물의 기능 분석미생물은 지구의 원자물질들의 생물지구화학적인 순환을 추진하는 역할을 한다. 미생물의 중요함과 방대한 다양성에도 불구하고, 미생물학에서 미생물의 분류는 간과 되고 있었다. 최근에 분자생물학적 방법과 안정한 동위원소를 생물표지자(biomarker)에 이용하는 방법을 결합하여 미생물들이 실제로 살고 있는 복잡한 군집에서 미생물의 분포와 기능을 연관지을 수 있게 되었다. Stable isotope probing(SIP)로 알려진 이 기술들은 안정한 동위원소가 표지된 기질을 미생물 군집으로 삽입하여 분석하고자 하는 분자들에서 동위원소의 표현을 뒤쫓아 기질의 운명을 추적하는 것이다. Stable Isotope probing은 방해가 최소화된 군집에서 기질 동화의 직접적 관찰을 가능하게 한다. 따라서 자연적 환경에서 미생물의 동정과 기능간의 연결고리를 위한 새로운 도구를 제시한다.
서 론
자연계에는 다양하고 수많은 미생물들이 존재하며 생명지구화학적 순환을 가능하게 하는 생분해에 핵심적인 역할을 하고 있다. 그러나 복잡하며 역동적인 군집에 존재하는 미생물들을 동정하고 행위를 조사할 수 있는 방법들은 제한 적이었다. 전통적으로 연구자들은 환경으로 부터 한가지 미생물을 추출하여 실험실에서 이 미생물의 유전자와 효소들을 연구하였다. 이런 접근법들은 미생물의 기작에 대한 많은 것을 밝혀 내었다. 또한 정교한 분석 화학적 방법들은 연구자들이 환경에서 아주 작은 수준의 수치 조차도 측정 할 수 있게 하였다. 그러나 실험실에서 연구되어진 하나의 미생물의 형질은 환경에서 유기체들과 군집을 이룰 때와는 다르며 토양 유기체 중 대부분이 실험실에서 배양되어진 적이 없다는 사실은 배양에 근거한 이러한 접근법의 위험성을 조명하고 있다(Giovanoni et al., 1990; Ward et al., 1990; Amann et al.,1992; Rappe and Giovannoni, 2003). 또 분자생물학적 방법이나 분석화학적 방법 하나만으로는 미생물 기작의 화학 반
