Abstract
Cycloheximide는 진핵새포 내의 단백질 번역과정에서 단백질을 합성을 억제하는 물질이다. Cycloheximide를 이용해 단백질 합성을 억제한 후 기존에 존재했던 단백질이 분해되는 시간을 측정함으로써 단백질 half-life를 결정할 수 있다. 단백질이 분해되는 과정은 lysosome-autophagy pathway, ubiquitin-proteosome pathway 두가지로 나눌 수 있다. Protein half-life는 단백질의 세포과정에서 단백질 풍부도를 결정하거나 질병에 특이적 단백질 degradation을 연구하는데 중요한 역할을 한다. 세포를 배양한 후, 단백질 농도를 결정하고 똑같은 양의 단백질을 취한다. 이후, SDS-page – transfer – western blotting 과정을 거쳐 결과를 확인한다. Western blotting 결과를 바탕으로 image J 프로그램을 이용해 단백질 양을 측정하고 시간에 따라 감소된 단백질 양을 확인해본다. cyclinD1과 -tubulin 단백질량의 백분율을 구하여 시간 경과에 따라 cyclin D1의 양이 감소한 것을 확인하였다. Cycloheximide의 기전을 이용하여 protein-half life를 결정할 수 있는 이유에 대해 생각해보았고 비슷한 실험을 진행한 논문 결과와 이번 실험의 결과를 비교해보고 차이가 나는 이유에 대해서도 생각해보았다.
Introduction
세포가 분열할 때 핵과 세포질을 포함하여 cell 내에서 진행되는 life cycle을 cell cycle 이라고 한다. 일반적으로 진핵세포의 cell cycle은 4단계(G1, S, G2, M)로 나눌 수 있다. G1기 에서는 cell의 크기가 커지기 시작하며 성장에 필요한 protein과 RNA를 만든다. 이 시기에서 cell 내의 DNA 함량은 변하지 않는다.
