Objective
• Development of a gradient porosity scaffold similar
to a filter made from collagen or fibrin
• Preparation of large open pores (20–50 um) at the scaffold surface to facilitate cell penetration, and dense pores (<1 um) at the scaffold bottom prevent cells leakage
• Evaluation of cell seeding efficiency, cell distribution and proliferation through the scaffold resulting in quantitative seeding in three dimensions

CONCLUSION
• A polarized membrane showed efficient cell seeding from results of SEM, histology, cell seeding experiments and proliferation studies.
• This study has a simple and reliable procedure, whereby scaffold materials can be processed into gradient membranes with good cell adhesion and seeding capabilities.
• The scaffold allows not only effective seeding but also the fast proliferation of the cells.
