IP의 원리는 그리 복잡하지 않다. 우리가 원하는 특정한 단백질 항원(specific target antigen)에 대항하는 항체(monoclonal 또는 polyclonal)를 cell lysate와 같은 샘플 안에 넣어주어서 그것의 타겟 항원과 면역 복합체(immune complex)을 형성하게 하는 것이다. 그런 후 그 면역 복합체는 Protein A 또는 Protein G가 붙어있는 고체 물질이 붙잡게 된다. 2. Rho GTPase & Actin Cytoskeleton RegulationRho GTPase는 Rho A, B, G를 유도하는 작은 GTP binding proteins의 Rho family의 멤버이다. Rho GTPase는 GTP-bound state에서는 활성을 띄지만, bound to GDP에서는 inactive을 나타낸다. Rho의 activation은 다양한 방면의 effector molecules과의 association을 나타내고 downstream signaling cascades의 activation을 이끈다. Rho proteins은 intrinsic GTPase activity을 나타낸다. Rho는 GTPase activating proteins (GAPs)에 의해 significantly stimulated되는 것이다.
이와 반대로, GTP exchange factors (GEFs)는 GDP의 GTP로의 전환을 자극하여 Rho를 activation시킨다. Rho GTPase는 actin cytoskeleton organization을 조절하고 assembly시킨다. 특히, actin stress fibers와 focal adhesion formation에 기여하는 바가 크며, cell spreading, migration, cytokinesis, morphogenesis와 같이 actin cytoskeleton에 의존하는 cellular processes에 관계가 깊다. Rho는 또한, gene transcription, G1 cell-cycle progression, cell-cell adhesion formation, and transformation을 포함하는 다른 pathways를 implicating한다고 알려져있다.
Actin cytoskeleton과 관련해서, Rho family GTPases는 actin cytoskeleton의 critical regulators이고 각각의 Rho GTPases는 specific actin reorganisation에 이르는 distinct signalling pathways을 activation을 한다고 알려져있다. 지금껏 동정된 23개의 mammalian Rho GTPases중에서 3개는 잘 characterised 되었다. Activated RhoA는 vinculin-rich focal adhesion complexes에 anchored되어 stress fibres라고 불리는 contractile actin-myosin filaments의 집합체를 자극한다.
